Semax

Semax (ACTH 4-10 Analog)

Peptide

Semax is a synthetic heptapeptide analog of ACTH 4-10 (Met-Glu-His-Phe-Pro-Gly-Pro) originally developed in Russia for cognitive enhancement, neuroprotection, and stroke recovery. It is primarily used to improve memory, focus, and mental clarity, and has clinical applications in Russia for ischemic stroke, optic nerve atrophy, and ADHD-like cognitive deficits.

Mechanism of Action

Semax increases brain-derived neurotrophic factor (BDNF) and nerve growth factor (NGF) expression in the hippocampus and cortex, promoting neuroplasticity and neuronal survival. It also modulates dopaminergic and serotonergic neurotransmission, enhances melanocortin receptor signaling, and inhibits enkephalin-degrading enzymes, prolonging endogenous opioid peptide activity that contributes to its anxiolytic and mood-stabilizing effects.

Dosing Protocols

nasal200-600mcg- Once or twice daily

Morning, or morning and early afternoon to avoid sleep disruption

Cycle: 4-8 weeks on, 2-4 weeks off

Most common route for self-administration. Standard Russian pharmaceutical preparation is 1% solution delivering ~50mcg per drop; typical dose is 2-4 drops per nostril. Reconstituted peptide solution should be refrigerated and used within 30 days.

injection200-500mcg- Once daily

Morning on an empty stomach or at least 1 hour before meals

Cycle: 4-8 weeks on, 2-4 weeks off

Subcutaneous injection provides higher bioavailability than nasal route. Used in clinical settings for neurological indications. Reconstitute with bacteriostatic water; store refrigerated.

Safety & Side Effects

Semax has a well-documented clinical safety record in Russia spanning over two decades, with no serious adverse events reported at therapeutic doses; however, long-term safety data outside of clinical settings remains limited. It is an unscheduled compound in most Western countries but is a prescription pharmaceutical in Russia, and purity/dosing accuracy of research-grade peptide vendors cannot be guaranteed.

Possible Side Effects

  • !Nasal irritation or mild burning upon intranasal administration
  • !Transient headache, particularly at higher doses
  • !Increased anxiety or overstimulation in sensitive individuals
  • !Fatigue or mild sedation in some users post-dose
  • !Appetite suppression
  • !Mild skin flushing when administered via injection
  • !Insomnia if dosed too late in the day
  • !Rare: irritability or mood dysregulation with prolonged use

Interactions

  • -May potentiate effects of stimulant medications (e.g., amphetamines, modafinil) due to additive dopaminergic activity — use with caution
  • -May enhance antidepressant effects of SSRIs or SNRIs through complementary serotonergic modulation — monitor for serotonergic symptoms
  • -Potential additive neuroprotective or cognitive effects when combined with racetams (e.g., piracetam) — not inherently dangerous but monitor response
  • -May lower cortisol response — use caution in individuals on corticosteroid therapy or with adrenal insufficiency
  • -No well-documented interactions with blood thinners or antihypertensives, but limited data warrants caution in polypharmacy contexts

Cost & Where to Buy

$30-$90
per month

Russian pharmaceutical nasal spray (Semax 1%) is inexpensive when imported but availability varies by country. Research-grade lyophilized peptide from Western vendors ranges higher; cost depends on dose frequency and vendor purity standards.
