Pregnenolone is an endogenous neurosteroid and steroid hormone precursor synthesized primarily from cholesterol in the adrenal glands, gonads, brain, and liver. It serves as the master precursor to virtually all steroid hormones including DHEA, progesterone, cortisol, estrogen, and testosterone. It is used as a supplement to support cognitive function, memory, mood, fatigue resistance, and as a precursor to downstream hormone production, particularly in aging individuals with declining steroid synthesis.
Mechanism of Action
Pregnenolone exerts effects via multiple mechanisms: it acts as a positive allosteric modulator of NMDA receptors and a negative modulator of GABA-A receptors, enhancing excitatory neurotransmission and promoting neuroplasticity in the brain. It also binds to microtubule-associated protein 2 (MAP2), supporting neuronal structure and function, and serves as the obligate precursor in the steroidogenesis pathway, converting to DHEA via CYP17A1 or to progesterone via 3β-HSD depending on tissue context. Dosing Protocols
Morning with meals or fat-containing food to enhance absorption
Cycle: Consider 5 days on, 2 days off or monthly breaks to avoid receptor desensitization
Most users start at 10-30mg and titrate up slowly. Doses above 50mg significantly increase conversion to downstream hormones and raise risk of androgenic or estrogenic side effects. Older adults may benefit from higher end of range.
