Pomegranate Extract
Pomegranate Extract (Punicalagins)
Pomegranate extract standardized to punicalagins is a polyphenol-rich supplement derived from the fruit rind and juice of Punica granatum. It is primarily used for cardiovascular support, antioxidant protection, and anti-inflammatory effects, with emerging research supporting benefits in exercise recovery, blood pressure regulation, and gut health. Punicalagins are among the most potent dietary antioxidants known, metabolized in the gut to urolithins which exert systemic effects.
Mechanism of Action
Punicalagins and their metabolites (ellagic acid, urolithins A and B) inhibit NF-κB signaling and downregulate pro-inflammatory cytokines including TNF-α, IL-1β, and IL-6. They also scavenge reactive oxygen species, upregulate endogenous antioxidant enzymes (superoxide dismutase, catalase), and inhibit aromatase and COX-2 enzymes. Additionally, pomegranate polyphenols modulate the gut microbiome to enhance urolithin production, which activates mitophagy pathways via PINK1/Parkin and supports mitochondrial quality control.

Dosing Protocols
With a meal
Look for extracts standardized to at least 40% punicalagins or 30% ellagic acid. Some clinical studies on blood pressure and exercise recovery used doses up to 1000mg daily. Higher doses (>2000mg/day) offer no clear added benefit and may increase GI side effects.
Safety & Side Effects
Pomegranate extract has a well-established safety profile in clinical studies at doses up to 1000mg daily for periods of up to 12 weeks; long-term safety data are generally reassuring but limited. Individuals on blood pressure medications or anticoagulants should exercise caution due to additive pharmacodynamic effects, and those with plant allergies should consult a healthcare provider before use.
Possible Side Effects
- !Gastrointestinal discomfort including bloating and loose stools at doses above 1000mg
- !Mild nausea, particularly when taken on an empty stomach
- !Allergic reactions in individuals sensitive to pomegranate or related fruits
- !Transient decrease in blood pressure, which may cause lightheadedness
- !Potential interference with iron absorption when taken concurrently with iron-rich meals
- !Rare reports of skin rash or itching in hypersensitive individuals
Interactions
- -May enhance anticoagulant effects of warfarin and other blood thinners — monitor INR closely
- -Additive blood pressure-lowering effect with antihypertensive medications — risk of hypotension
- -May inhibit CYP3A4 and CYP2C9 enzymes, potentially raising plasma levels of statins, calcium channel blockers, and certain immunosuppressants
- -May have additive effects with other antioxidant supplements (e.g., resveratrol, quercetin) — generally safe but monitor for hypotension
- -Co-administration with ACE inhibitors may produce excessive blood pressure reduction
Cost & Where to Buy
Price varies significantly based on standardization percentage and brand quality. Bulk powders are cheapest; standardized capsules from reputable brands (NOW, Life Extension, Jarrow) typically run $15-35/month. Avoid unstandardized products with no punicalagin content listed.
