L-Leucine is an essential branched-chain amino acid (BCAA) that cannot be synthesized by the human body and must be obtained through diet or supplementation. It is primarily used to stimulate muscle protein synthesis, support recovery from resistance exercise, and preserve lean muscle mass during caloric restriction or aging. Leucine is considered the most anabolic of the three BCAAs and is a key trigger for muscle-building signaling pathways.

Mechanism of Action

Leucine activates the mammalian target of rapamycin complex 1 (mTORC1) signaling pathway, which serves as a primary regulator of muscle protein synthesis and cellular growth. It acts as a nutrient sensor, directly binding to Sestrin2 and the GATOR2 complex to activate mTORC1, stimulating downstream effectors S6K1 and 4E-BP1 to initiate ribosomal translation of muscle proteins. Leucine also enhances insulin secretion from pancreatic beta cells and suppresses protein degradation via the ubiquitin-proteasome pathway, creating a net anabolic environment in skeletal muscle.

A minimum of 2-3g per dose appears to be the threshold required to maximally stimulate mTORC1 signaling. Older adults (>65 years) may benefit from the higher end of the range (3-5g per dose) due to anabolic resistance. If consuming whey protein or a complete protein meal, additional leucine supplementation may offer limited benefit as dietary protein already provides leucine.
