GHRP-6

Growth Hormone Releasing Peptide-6

Peptide

GHRP-6 is a synthetic hexapeptide that stimulates the pituitary gland to release growth hormone (GH) in a pulsatile, physiological manner. It is primarily used in research and off-label contexts for bodybuilding, anti-aging, recovery from injury, and improving body composition by increasing GH and downstream IGF-1 levels. Unlike exogenous HGH, GHRP-6 works by amplifying the body's own GH secretion, preserving the natural feedback loop.

Mechanism of Action

GHRP-6 acts as a ghrelin mimetic, binding to the growth hormone secretagogue receptor 1a (GHSR-1a) in the pituitary and hypothalamus, triggering a strong and rapid GH pulse. It simultaneously suppresses somatostatin (the GH-inhibiting hormone), further amplifying GH release. The resulting GH pulse stimulates hepatic IGF-1 production, which mediates many of the downstream anabolic, lipolytic, and recovery-promoting effects.

Dosing Protocols

injection100-300mcg- 2-3x daily

On a fully empty stomach (2-3 hours after last meal, ideally upon waking, pre-workout, and before bed)

Cycle: 12 weeks on, 4 weeks off

Administer subcutaneously into abdominal fat; insulin syringes (29-31 gauge) are standard. GH response desensitizes with more than 3 doses per day. Pairing with a GHRH analog (e.g., CJC-1295 or Mod GRF 1-29) at the same injection time produces a synergistic and significantly larger GH pulse. Avoid carbohydrates and fats within 30-60 minutes of injection to prevent blunting of the GH response.

Safety & Side Effects

GHRP-6 has a relatively well-tolerated short-term safety profile in healthy adults, but long-term human safety data is limited as most evidence comes from animal studies and anecdotal reports. It is an unscheduled research chemical in most jurisdictions but is not approved for human use by regulatory agencies such as the FDA; purchase and use exist in a legal gray area, and quality control across peptide vendors is highly variable.

Possible Side Effects

  • !Intense, acute hunger and appetite stimulation within 20-30 minutes of injection (most common and pronounced side effect)
  • !Water retention and mild peripheral edema due to elevated GH and IGF-1
  • !Elevated cortisol and prolactin levels, particularly at higher doses
  • !Lethargy and fatigue, especially following the GH pulse (most notable after evening doses)
  • !Injection site redness, irritation, or mild bruising
  • !Tingling or numbness in the extremities (carpal tunnel-like symptoms with prolonged use)
  • !Potential for decreased insulin sensitivity and transient hyperglycemia with chronic use
  • !Headache, particularly in early weeks of use as GH levels increase

Interactions

  • -Insulin and hypoglycemic agents: GHRP-6 can impair insulin sensitivity and raise blood glucose, potentially requiring dose adjustments in diabetics or those using insulin
  • -Corticosteroids: May blunt the GH-releasing effect of GHRP-6 and compound HPA axis dysregulation due to GHRP-6's own cortisol-elevating properties
  • -Thyroid hormone (T4/T3): Elevated GH and IGF-1 increase peripheral conversion of T4 to T3; individuals on thyroid replacement therapy may need monitoring
  • -Exogenous HGH: Concurrent use amplifies GH excess risk, including increased incidence of edema, carpal tunnel symptoms, and insulin resistance
  • -Dopamine agonists (e.g., cabergoline): May partially offset GHRP-6-induced prolactin elevation; sometimes used adjunctively for this purpose

Cost & Where to Buy

$30-$90
per month

Pricing reflects research-grade peptide vendors; a 5mg vial typically costs $15-$35 and covers approximately 16-50 injections at 100-300mcg per dose. Monthly cost varies significantly with dosing frequency, dose size, and vendor. Higher-purity, third-party tested sources command a premium. Often purchased alongside GHRH peptides, increasing total monthly spend.
