Coleus forskohlii (Forskolin)
Forskolin is a labdane diterpene compound extracted from the root of Coleus forskohlii, a plant in the mint family. It is primarily used for body composition support, testosterone optimization, and as a research tool for studying cAMP-mediated signaling. Clinically, it has been investigated for asthma, glaucoma, and cardiovascular conditions.
Mechanism of Action
Forskolin directly activates adenylyl cyclase, the enzyme responsible for converting ATP to cyclic AMP (cAMP), thereby bypassing receptor-level signaling and broadly elevating intracellular cAMP concentrations across multiple tissue types. Elevated cAMP activates protein kinase A (PKA), which phosphorylates downstream targets including hormone-sensitive lipase (promoting lipolysis), thyroid hormone synthesis pathways, and Leydig cell testosterone production via StAR protein upregulation. This mechanism also causes smooth muscle relaxation, explaining its bronchodilatory and vasodilatory effects.
