CDP-Choline
Citicoline (CDP-Choline)
CDP-Choline (citicoline) is a naturally occurring nucleotide that serves as an intermediate in the biosynthesis of phosphatidylcholine, a major component of neuronal cell membranes. It is widely used as a nootropic to enhance cognitive function, memory, and mental clarity, and is also clinically investigated for stroke recovery and age-related cognitive decline. Upon ingestion, it provides both choline and cytidine (which converts to uridine), supporting acetylcholine synthesis and neuronal membrane integrity.
Mechanism of Action
CDP-Choline is hydrolyzed in the gut and liver into choline and cytidine; choline crosses the blood-brain barrier where it serves as a precursor for acetylcholine synthesis via choline acetyltransferase, enhancing cholinergic neurotransmission. Uridine derived from cytidine supports phosphatidylcholine synthesis and potentiates dopaminergic signaling by increasing striatal dopamine receptor density. Additionally, citicoline upregulates mitochondrial ATPase activity and reduces free radical generation, providing neuroprotective effects.

Dosing Protocols
Morning and early afternoon, with or without food
Most clinical trials used 500mg twice daily (1000mg/day total) for therapeutic effects. For general cognitive enhancement, 250-500mg once daily may be sufficient. Avoid dosing in the evening as it may interfere with sleep due to increased cholinergic and dopaminergic activity.
Safety & Side Effects
CDP-Choline has an excellent safety profile and is well-tolerated in the majority of clinical trials, including studies in elderly and stroke populations at doses up to 2000mg/day. It is an over-the-counter supplement in the United States but is a prescription medication (Ceraxon, Somazina) in several European and Latin American countries; individuals with a history of hypertension or bipolar disorder should exercise caution and consult a physician before use.
Possible Side Effects
- !Headache, particularly at doses above 1000mg/day in choline-sensitive individuals
- !Insomnia or difficulty falling asleep if taken in the evening
- !Gastrointestinal discomfort including nausea and diarrhea at higher doses
- !Increased blood pressure reported in some users at doses above 1000mg/day
- !Restlessness or overstimulation, especially when combined with other cholinergic compounds
- !Muscle tension or jaw tightness due to elevated acetylcholine levels
Interactions
- -May potentiate cholinergic effects of acetylcholinesterase inhibitors (e.g., donepezil, rivastigmine), increasing risk of excessive salivation, nausea, and bradycardia
- -May modestly increase blood pressure - use caution when combined with antihypertensive medications as dosage adjustments may be needed
- -Additive cognitive and cholinergic effects when combined with other choline sources (Alpha-GPC, choline bitartrate) - reduce individual doses to avoid cholinergic overload
- -May reduce effectiveness of meclofenoxate (centrophenoxine) due to overlapping cholinergic mechanisms
- -Synergistic nootropic effects reported with racetams (piracetam, aniracetam), which increase acetylcholine turnover and may deplete choline stores
Cost & Where to Buy
Generic CDP-Choline powder or capsules from reputable supplement brands (Jarrow, Cognizin) typically cost $15-30/month at 500mg/day; branded Cognizin-certified products command a premium of $30-45/month. Prescription formulations outside the US can be significantly more expensive.
